Sik3 Suppresses Disease in Murine OA

Risk factors for osteoarthritis include age, which is associated with thinning of articular cartilage. We generated chondrocyte-specific salt-inducible kinase 3 (Sik3) conditional knockout mice that were resistant to osteoarthritis with thickened articular cartilage. We also identified a compound, pterosin B, as a Sik3 pathway inhibitor. Either Sik3 deletion or intraarticular injection of mice with pterosin B inhibited chondrocyte hypertrophy and protected cartilage from osteoarthritis. Our results suggest Sik3 is a target for treatment of osteoarthritis, with pterosin B as a candidate therapeutic.
